Hyperosmotic stress-induced somatic embryogenesis and its continuous culture in Japanese honewort (<i>Cryptotaenia japonica</i>)

چکیده
Japanese honewort (Cryptotaenia japonica) is consumed as a traditional vegetable and has medicinal applications. In Japan, C. japonica mainly produced using hydroponic culture systems; however, damping-off often caused by the adherence of pathogens to its seeds. Therefore, use sterile artificial seeds in likely be effective for preventing disease. this study, we established methods stress-induced somatic embryogenesis seed production honewort. Shoot apex explants from seedlings were treated with 0.7 M sucrose hyperosmotic stress 3 or 6 weeks, then transferred stress-free conditions. Somatic embryos formed after conditions 7 weeks. Stress-treated shoot that cultured Murashige Skoog liquid medium shaking. After 2 weeks culture, approximately 800 each explant. continuously during 37 under same Thus, was effectively induced via stress, embryogenic competence maintained stress- phytohormone-free The used produce enveloping whipped alginate gel avoid hypoxic had high germination rate (72%). This system suitable sterile, highly productive

Somatic Embryogenesis and Efficient Plant Regeneration in Japanese Cypresses
There are six species in the genus Chamaecyparis worldwide, of which two, namely the Hinoki cypress (Chamaecyparis obtusa Sieb. et Zucc.) and Sawara cypress (Chamaecyparis pisifera Sieb. et Zucc.) are distributed in Japan (Maruyama et al., 2002). The Hinoki cypress is one of the most important commercial timber trees in Japan, representing about 25% of the plantation area in the country. Howeve...

Synchronization of somatic embryogenesis was achieved in a carrot (Daucus carota L. cv. "Kurodagosun") suspension culture by sieving the initial heterogeneous cell population, by density gradient centrifugation in Ficoll solutions, and by subsequent repeated centrifugations at a low speed (50g) for a short time (5 seconds), followed by transferring the cell clusters obtained, which were compose...

متن کاملZygotic embryogenesis versus somatic embryogenesis
This review will summarize molecular and genetic analyses aimed at identifying the mechanisms underlying the sequence of events during plant zygotic embryogenesis. These events are being studied in parallel with the histological and morphological analyses of somatic embryogenesis. The strength and limitations of somatic embryogenesis as a model system will be discussed briefly. The formation of...
متن کاملSomatic Embryogenesis
Somatic embryogenesis is an asexual form of plant propagation in nature that mimics many of the events of sexual reproduction. Also, this process may be reproduced artificially by the manipulation of tissues and cells in vitro. Some of the most important factors for a successful plant regeneration are the culture medium and environmental incubation conditions. In vitro somatic embryogenesis is ...
